韓国科学技術院(KAIST)は、KAISTと蔚山科学技術院(UNIST)の研究者らが、膵臓がんの微小環境障壁を克服する形状変化型デバイスを開発したと発表した。研究成果は学術誌Advanced Materialsに掲載された。KAISTのイ・キョン・ジェ(Keon Jae Lee)教授(左)、UNISTのクォン・テヒョク(Tae-Hyuk Kwon)教授(左から2人目)と研究チームメンバーら従来の膵臓がん治療は、腫瘍微小環境(TME)という大きな障害に直面している。この生物学的障壁はがん細胞を包み込み、化学療法薬や免疫細胞の浸潤を大きく制限する。一方、光線力学的療法(PDT)は膵臓がん治療の有望な代替手段となるが、レーザーなどの既存の外部光源は深部組織を効果的に貫通できず、健康な臓器への熱損傷や炎症のリスクを伴う。これらの課題を解決するため、KAISTのイ・キョン・ジェ(Keon Jae Lee)教授の研究チームはUNISTのクォン・テヒョク(Tae-Hyuk Kwon)教授と共同で、深部組織に効果的に光を届ける埋め込み可能な形状変化型3DマイクロLEDデバイスを開発した。この技術の鍵は、タコのような柔軟な構造にあり、膵臓の腫瘍全体を包み込むことができる。デバイスは腫瘍の生理的な拡大や収縮に関わらず、正常な組織を保持し、がん細胞を標的とした均一で連続的かつ低強度の光刺激を可能にする。マウスを用いた生体実験では、このデバイスによる顕著な治療効果が示された。これはわずか3日間の実験であるにも関わらず、PDTの限界を克服し、腫瘍線維組織を64％減少させ、膵臓組織を正常組織へ回復させた。イ教授は、「この研究は、膵臓がん治療における最大の障害であるTMEを直接破壊することで、新たな治療パラダイムを提示しました」と述べ、研究の今後について、「この技術を人工知能(AI)と統合したスマートプラットフォームへ発展させ、リアルタイムの腫瘍モニタリングと個別化された治療を可能にすることを目指しています。現在、臨床試験とヒトへの応用に向けた商業化を進めるパートナーを探しています」と説明した。Overall concept of 3D Shape-morphing micro-LEDs (SMLEDs).
